It means both. If it is said as a question, “Let us play?” or “Will you let us play?” it means allow us to play.
If it is said as a statement, “Let us play” or “Let’s play” it means come and play.

In NEITHER CASE is it a question, so it is NEVER written with a “?”. You, however, are asking us a question, so YOUR question mark is required, and it is in the proper place, OUTSIDE the quotes.
