I love Hairspray!
A 45 was a type of vinyl record (the big black ones) that was of great quality in the sixties. Loads of kids would’ve just died to have a good 45 versus a 33. It was like the Iphone of their generation, lol.

Back in the day, before Cd’s, they made vinyl records. When played, they played at different speeds. For example, a full album would usually play at 33 rpm (revolutions per minute). A 45 was a smaller record which usually was a single (actually 2 singles — one on each side). As you might have guessed these smaller records played at 45rpm. People used to call them 45’s — today you would call them “singles”.
To answer your question about what that line means — it means that life without love is like living w/o a hit single that you want. You know it’s out there and you want it, but you don’t have it.
